By RAW, they each threaten adjacent squares only, not the square that they are in. So a sentinel couldn't attack the medium creature in his square, but everyone around him could. A Large creature, however, occupies adjacent squares as well as the ones that the sentinels are in, so that wouldn't be a problem for the sentinels.
